摘 要:目的:确定骨髓间充质干细胞(BMSCs)在异种胸腺内的演化过程,为进一步进行BMSCs胸腺修饰诱导异种免疫耐受的研究提供可行性依据。方法:取豚鼠BMSCs经分离纯化体外扩增至第3代,并行胎牛血清-Fe2O3和4,6-二咪基-4-联苯基吲哚(DAPI)标记,标记后的豚鼠BMSCs行胸腺注射,7天后进行切片普鲁士蓝-中性红染色和荧光显微镜观察豚鼠BMSCs演变过程。结果:BMSCs移植至异种胸腺中能够生存,并能转化为多种组织细胞成分,即仍具有多向分化能力。结论:异种BMSCs胸腺修饰在诱导异种受体移植免疫耐受方面将是可行的。Objective:To observe the proliferation and differentiation of xenogeneic bone mesenchymal stem cells(BMSCs) in the thymus and to explore the feasibility of intrathymic inoculation with xenograft-BMSCs in inducing immunotolerance.Methods:BMSCs of guinea pigs were cultured in medium to P3,and labeled with fetal bovine serum-Fe2O3 and 4,6-diamidino-2-phenylindole(DAPI).The marked BMSCs were inoculated into thymuses gland of rats.The thymuses were sliced up and dyed with perls solution seven days later and observed directly under the fluorescenc microscope.Results:BMSCs survived long term in the micro-environment of the xenogeneic thymuses,and had the ablitity to differentiate into various cell types.The xenogeneic BMSCs had the capablilty of multiple rounds of division.Conclusion:It is feasible to induce immune tolerance by intrathymic inoculation with xenograft-BMSCs.
