摘 要:目的:探讨大块异种脱细胞骨基质的制备及生物相容性,为其在骨缺损修复领域中的临床应用提供实验依据。方法:取牛股骨下端松质骨切割制作4.0 cm×2.0 cm×1.0 cm大小的骨块,用10%NaCl和0.5%曲拉通X-100联合脱细胞处理,分为48 h、72 h、96 h三个时间段;处理后的大块异种脱细胞骨基质行HE染色、Masson染色及扫描电镜观察,观察大块异种脱细胞骨基质的脱细胞程度;用处理后的大块脱细胞骨基质制作浸提液,行急性毒性实验、热源实验,观察大块异种脱细胞骨基质有没有毒性反应及热源性;用处理后的大块异种脱细胞骨基质材料与SD大鼠骨髓间充质干细胞体外复合培养,观察细胞生长、细胞增殖及蛋白质含量测定。结果:4.0 cm×2.0 cm×1.0 cm骨块经过10%NaCl和0.5%曲拉通X-100联合处理72 h组及96 h组的异种脱细胞骨基质经HE染色及扫描电镜观察未见细胞成分,Masson染色观察胶原纤维基本结构未被破坏,材料浸提液急性毒性实验、热源实验符合规定标准,且骨髓间充质干细胞能够贴附在异种脱细胞骨基质孔隙内生长,细胞生长及蛋白合成功能不受异种脱细胞骨基质的影响。结论:大块异种脱细胞骨基质具有良好的生物相容性,可作为骨缺损修复的支架材料。Objective: To explore the best methods of preparation of massive heterogenous acellular bone extracellular matrix and its biocompatibility,to provide experimental basis for the clinical use in the bone reconstruction field.Methods:Bovince lower femur cancellous bone was cut into 4.0 cm×2.0 cm×1.0 cm,then was approached acellularly by 10% sodium chloride and 0.5% TritonX-100 with three time periods of 48 h、72.96 h、Specimens were stained with HE,Masson and observed with naked eyes,microscopy and scanning electron microscopy,to observe the acellular degree of ABECM;To evaluate the biocompatibility through acute toxicity test and pyrogen test;Bone marrow mesenchymal stem cells were seeded into acellular bone extracellular matrix and cultured in vitro,then their morphological characteristics,growth curve and proliferation,protein content were detected.Results:There were no cellular components in respectively histological slides of the bones,but the basic structure of extracellular matrix existed in 4.0cm×2.0cm×1.0cm with 72h and 96h.Massive heterogenous acellular bone extracellular matrix had no toxicity and did not contain the pyrogenic material.Bone marrow mesenchymal stem cells could attach to the inner surface of acellular bone extracellular matrix,and their capability of protein production were not affected by it.Conclusion:Massive heterogenous acellular bone extracellular matrix can be used as scaffold with good biocompatibility in the repair and treatment for bone defects.
