摘  要:目的:研究薯蓣皂苷对正常大鼠心室肌细胞L-钙电流(ICa,L)的影响,旨在探讨薯蓣皂苷对心肌缺血再灌注(I/R)损伤保护作用的可能机制。方法:酶解法分离大鼠单个心室肌细胞,应用全细胞膜片钳技术记录薯蓣皂苷低(10μg/ml)、中(20μg/ml)、高(30μg/ml)三个剂量对大鼠单个心室肌细胞ICa,L的影响。结果:薯蓣皂苷10μg/ml不影响ICa,L峰值,而20μg/ml和30μg/ml能使ICa,L峰值降低,抑制呈剂量依赖性;20μg/ml和30μg/ml薯蓣皂苷使I-V曲线上移,内向电流减小,但基本不改变曲线的形状。30μg/ml薯蓣皂苷使失活曲线向负电位方向变化,曲线左移,各剂量均对激活曲线无影响。结论:薯蓣皂苷通过促进ICa,L失活的过程抑制正常大鼠心室肌细胞I,减小I峰值,这可能是其抗缺血再灌所致心律失常的作用机制之一。Aim: To investigate the effect of dioscin on L-type calcium current(I Ca,L) of ventricular cardiomyocytes and explore the possible protective mechanism of dioscin against the ischemia/reperfusion(I/R) injury.Methods: Whole-cell patch clamp was used to record I Ca,L in single ventricular cardiomyocytes of rats.Results: In ventricular cardiomyocytes of rats,calcium peak current was decreased by 20~30μg/mldioscin in a concentration-dependent manner,while 10μg/ml had no effect on it.The current-voltage curve was shifted upwards by 20~30μg/mldioscin,which curve shape had no significant changes;The inactivation curve was shifted to more negative potentia1 by 30μg/ml dioscin.but the activation curve was not altered.Conclusion: Dioscin inhibits I Ca,L of ventricular cardiomyocytes by facilitating the inactivation of I Ca,L,which may partly attribute to the antiarrhythmic action in ischemia/reperfusion(I/R) injury.
