摘 要:目的:探讨短间隔高压氧预处理对大鼠脊髓缺血再灌注损伤后NF-κB表达的影响。方法:80只成年雄性Wistar大鼠,随机分为2组(每组n=40):对照组为常压空气组;HBO组为短间隔高压氧预处理组。采用肾下腹主动脉阻断法造成脊髓缺血再灌注损伤,观察两组再灌注后4 h、12 h、24 h、48 h时的后肢运动神经功能评分;再灌注48 h时取出腰段脊髓组织(L5-7)行石蜡包埋、切片,采用免疫组织化学法检测各组动物脊髓组织中NF-κB表达变化。结果:再灌注4 h、12 h、24 h、48 h时,HBO组神经功能学评分均明显优于对照组(P<0.05)。HBO组各时间点NF-κB阳性表达均明显低于对照组。HBO组和对照组后肢运动神经功能学评分与脊髓组织NF-κB表达呈正相关(r=0.72,0.65,P<0.05)。结论:高压氧预处理对缺血再灌注损伤的脊髓具有保护作用,且其所诱导的脊髓缺血耐受的机制与NF-κB有关。AIM:To investigate the effect of hyperbaric oxygen preconditioning on the expression of NF-κB in rats spinal cord tissue after ischemia-reperfusion.METHODS: 80 male rats were randomly allocated to 2 groups(40 in each group): animals in control group received room air and normal ambient pressure;animals in HBO group received HBO pretreatment.At 4,12,24,48 h after reperfusion,the neurologic status was scored by the Tarlov criteria.All the animals were sacrificed at 48 h after reperfusion and their spinal cords(L5-7)were removed to determine expression of NF-κB by immunohistochemistry.RESULTS: The neurologic function scores in HBO group were higher than that in control groups at 4,12,24,48 h after reperfusion(P0.05).The expression of NF-κB in HBO group were higher than that in control group at 4,12,24,48 h after reperfusion(P0.05).A positive correlation was found between the neurologic function scores and the expression of NF-κB in spinal cord(r=0.72,0.65,P0.05).CONCLUSION: Hyperbaric oxygen preconditioning can significant1y improve the motor function of hind limbs in rats after spinal cord ischemia-reperfusion injury,and it might be re1ated with inhibition of the expression of NF-κB.
