摘  要:目的比较狄氏剂对不同发育阶段的人多巴胺能SH-SY5Y细胞的毒性。方法以人多巴胺能SH-SY5Y细胞为模型,用10μmol/L全反式视黄酸(all-trans-retinoic acid,RA)分别诱导分化0、4和7 d,以1、3、10、30、100μmol/L的狄氏剂对细胞染毒24 h,以MTT法测定细胞存活率;按5×103个/孔的密度将细胞传至96孔板,用10μmol/L RA分别诱导分化0、4和7 d,以10、30、100μmol/L的狄氏剂对细胞染毒24 h,细胞活性/细胞毒性(LIVE/DEAD)试剂盒检测细胞的存活率和绝对死亡率。结果 MTT试验结果显示,狄氏剂可引起SH-SY5Y细胞存活率呈剂量依赖性下降(均P<0.01),与未分化(0 d)和已分化(7 d)的细胞相比,分化中(4 d)的细胞存活率在较低剂量(3μmol/L)就有明显下降(P<0.05)。细胞活性/细胞毒性染色结果显示,10~100μmol/L的狄氏剂可产生明显细胞毒性,SH-SY5Y细胞的存活率呈剂量依赖性降低,绝对死亡率呈剂量依赖性增高(均P<0.05)。已分化的细胞较未分化细胞的绝对死亡率更高(P<0.05),而分化中的细胞又比已分化细胞的绝对死亡率更高(P<0.05)。结论狄氏剂对不同发育阶段的多巴胺能神经细胞均有毒性作用,且分化中的细胞对狄氏剂的毒性作用更敏感。Objective To compare the cytotoxieity of dieldrin on dopaminergic neurons at different developmental stages. Methods All-trans-retinoic acid (10 μmol/L) was used to induce differentiation of SH-SYSY cells. MTr and Live/Dead viability/ cytotoxicity kit were used to assay the cytotoxicity of dieldrin to the undifferentiated (0 d),differentiating (4 d) or differentiated (7 d) SH-SY5Y cells. Results The results of MTT showed that cells viability was decreased by dieldrin in a dose depended manner, The differentiating SH-SYSY cells were most sensitive to dieldrin. The results of LIVE/DEAD also showed that cells viability was decreased by dieldrin in a dose-dependent manner, The cells exposed to dieldrin at concentrations of 10-100 μmol/L were induced dose-dependent cell death. The differentiating/differentiated SH-SY5Y cells were more sensitive to dieldrin than the undifferentiated cells, and the differentiating SH-SYSY cells were more sensitive than the differentiated cells. Conclusion The dieldrin can induce cytotoxicity to dopaminergie neurons of different developmental stages,and the differentiating cells are most sensitive.
