摘 要:建立了分离测定槐角提取液中染料木素的在线酸坝富集-毛细管区带电泳方法,考察了酸的种类、酸的及浓度、进样/进酸时间比例等因素对堆积效率的影响。实验以20 mmol/L硼砂(pH10.5)作为缓冲溶液,200 mmol/L柠檬酸溶液(pH1.7)作为酸坝,进样、进酸时间分别为180s和45s,在分离电压15kV,检测波长254 nm的条件下,染料木素5 min内出峰,检出限可达10 nmol/L,富集倍数为300多倍。A method was established for analysis based on online acid bahage stacking. The of genistein in Fructus Sophorae by capillary zone electrophoresis effect of several factors (such as acid type, acid pH, acid concentration and the ratio of injection time of sample and acid) on stacking efficiency was investigated. Genistein was detected within 5 nfin under the following conditions:20 mmol/L borax (pH 10.5) as running buffer, 200 mmol/L citric acid(pH 1.7) as acid barrage, injection time of 180s for sample, 45s for acid, separation voltage of 15 kV, detection wavelength 254 nm. In this method, the limit of detection for genistein was 10 nmol/L with more than 300 fold accumulation concentration.
