摘 要:建立了一种人体尿液中尿酸含量的高效液相色谱测定方法。采用ACE5AQ亲水色谱柱,pH3.2的乙酸水溶液为流动相,检测波长280nm。尿酸含量在7.1~224.6μg∥mL范围内线性关系良好,平均加样回收率为99.7%~100.5%,RSD小于1.4%。将该法用于健康人和肝硬化病人尿液样本的测定,两类样本中尿酸含量无显著性差异。A HPLC method was developed for the determination of uric acid in human urine. Urine samples were successfully separated on an ACE 5 AQ hydrophilic column with mobile phase of acetic acid solution (pH3.2).The detection wavelength was set at 280 nm. The linear curve of uric acid concentration was within the range of 7.1 -224.6 pμg/mL and the average spiked recoveries were 99.7% - 100.5% with the RSD below 1.4%. The analysis results of urine samples showed that there was no significant difference in urinary uric acid concentration levels between healthy persons and liver cirrhosis patients. The established HPLC method is simple, fast, accurate, and reliable, and thus can be applied to the determination of urle acid in ,clinical urine samples.
