全新结构抗肿瘤药物TNBG对裸鼠移植瘤生长抑制作用及药物毒副作用初步评价

摘　　要：目的观察TNBG对人肝癌细胞株QGY-7701裸鼠移植瘤生长抑制情况,初步评估药物的毒副作用。方法采用裸鼠复制人肝癌移植瘤模型。实验分对照组和给药组,腹腔注射给药,持续5周。治疗期间定期测量肿瘤大小,观察裸鼠生存状况。实验结束时处死裸鼠,测量肿瘤体积计算抑瘤率;眼眶取血,分离血清检测血脂、肝、肾功能;摘取裸鼠肿瘤及主要脏器组织作苏丹Ⅲ染色观察脂质沉积情况。结果 TNBG对移植瘤的抑瘤率为60.1%;各实验组裸鼠血脂、肝、肾功检测与对照组比无显著性差异(P>0.05);苏丹Ⅲ染色显示给药组裸鼠主要脏器组织内无脂质沉积,而肿瘤组织中可见大量脂质沉积。结论 TNBG对人肝癌细胞裸鼠移植瘤有较明显的抑制作用,毒副作用小,抗肿瘤作用具有选择性。Objective To examine the antitumor efficacy of TNBG on hepatocellular carcinoma （ QGY-7701 ） cells which was inoculated in the athymie nude mice, and to evaluate the side effects of TNBG. Method Nude mice were used to duplicate the xenogtafts models of human hepatocareinoma cells, divided into control group and TNBG group. The TNBG group was administrated with TNBG by intraperitoneal injection which lasted for 5 weeks. To compare the therapeutic effects, the tumor size and survival condition of nude mice were recorded during the administration. At the end of experiment ,the tumors were dislodged to calculate the inhibition rate; the serum was separated to detect the bloodfat,liver function and renal function; the main tissues of nude mice were removed and fixed in folanalin to observe the fatty substance by sudan Ⅲ stain. Results The tumor inhibition rate was 60. 1%. There were no obvious differences between the control group and TNBG group on the levels of serum blood-fat and the levels for evaluating the liver function and renal function. In comparison with control group, there were no fatty deposition in main tissues and tumor tissue of control group except in the tumor tissue of TNBG group. Conclusion TNBG had good antineoplastic effects and small side effects on xenogtafts of human hepatocarnoma cells in nude mice. the anti-tumor effects had somewhat selectivity.
