摘  要:目的应用红外可视脑片膜片钳技术观察精氨酸加压素对大鼠视前区-下丘脑前部(PO/AH)温度敏感神经元放电活动的影响。方法应用红外微分干涉相差技术和膜片钳技术实时观察57只雄性SD大鼠下丘脑PO/AH区神经元形态,鉴定温度敏感性。精氨酸加压素(AVP)和V1a受体阻断剂胞外给药,观察对温度敏感神经元放电活动的影响。结果红外可视条件下,下丘脑PO/AH区神经元的胞体清晰可见。AVP引起热敏神经元放电频率升高(P<0.01),冷敏神经元放电频率下降(P<0.01);V1a受体阻断剂能降低热敏神经元的放电频率(P<0.01)和提高冷敏神经元的放电频率(P<0.05)。结论 AVP通过V1a受体影响PO/AH区温度敏感神经元的放电活动,而引起体温降低。Objective To observe the effect of arginine vasopressin(AVP)on firing activities of rat temperature sensitivity neurons in preoptic-anterior hypothalamus area(PO/AH)slice recorded by infrared visual patch clamp technique.Methods On PO/AH area slices of 57 SD male rats,the shapes of neurons in PO/AH were observed in real-time by infrared differentiate interfere difference technique.The spontaneous firing were recorded by whole-cell patch clamp technique.Firing activities were influenced by temperature,AVP and V1a receptor antagonist in ACSF.Result Under the infrared visua1 patch clamp,the neurona1 bodies in PO/AH were very clear.AVP increased the firing rate of warm-sensitive neurons(P0.01)and decreased the firing rate of cold-sensitive neurons(P0.01).On the contrary,V1a receptor antagonist reduced the firing rate of warm-sensitive neurons(P0.01)and raised the firing rate of cold-sensitive neurons(P0.05).Conclusions AVP affect firing activities of temperature sensitivity neurons in PO/AH by V1a receptor.It has some relationship with the phenomenon that AVP reduced normal temperature.
