摘 要:从病猪肿大的跗关节中分离了1株疑似肠球菌菌株,经过常规方法进行染色特性、培养特性观察以及敏感性和致病试验,利用Vitek-32全自动细菌鉴定系统进行生化特定,并用PCR方法扩增分离株的16S rRNA基因克隆测序,与GenBank上登录的相关菌株及3个粪肠球菌标准菌株进行16S rRNA序列比较、同源性分析并构建系统发育树.结果表明,该分离株形态及染色特性与肠球菌一致,对仔猪具有一定的致病性,并对临床常用的7种药物产生了耐受性;Vitek-32生化鉴定和16S rRNA基因同源性分析及比对结果均显示其为粪肠球菌.An enterococcus-like isolated was isolated from piglet with lesion tarsal jiont. Morphological and cultural characteristics were observed, and drug sensitivity and piglet challenge tests were performed using the conventional approaches. Biochemical tests for the isolate were carried out by the Vitek-32 automatic microorganism identification system. Moreover, 16S rRNA genes of the isolate were amplified by means of polymerase chain reaction (PCR) and the target fragments were cloned and sequenced. A dendrogram was constructed by the MegAlign of DNA Star version 5. 0 after the sequences of 16S rRNA gene were compared with the related enterococci reported on GenBank. The results showed that the isolate was coherent to Enterococcus in morphological and cultural characteristics and was pathogenic to piglets. The results of drug sensitivity tests showed that the isolate was resistant to 7 clinically used antibiotics. Both Vitek-32 identification and 16S rRNA gene analysis results confirmed that the isolate was Enterococcus faecalis.
