摘 要:应用流式细胞术( F C M )检测分析了双氢青蒿素对杜氏利什曼原虫前鞭毛体 D N A 的作用。4 个不同浓度(177×10- 4,353×10- 4,71×10- 4,141×104m ol/ L)的药物对虫体 D N A 均有抑制作用,用药组虫体 D N A 含量的荧光分布随着药物浓度的升高而减弱,抑制率分别为 323% ,326% ,864% ,892% ,与对照组相比有显著性差异( P< 005)。后两个用药组与前两个用药组抑制率有显著性差异( P< 0005)。本研究结果与光镜观察、3 H Td RIn the paper, the effect of dihydroartemisinin on Leishmania donovani promastigotes DNA is analysed by flow cytometry. L.donovani promastigotes DNA was inhibited by dihydroartemisinin under four different concentrations (1.77×10 -4 , 3.53×10 -4 , 7.1×10 -4 , 14.1×10 -4 mol/L). In experiment groups, the fluorescent distribution of DNA content in L.donovani promastigotes decreased when drug concentration increased and with inhibition rates of 32.3%, 32.6%, 86.4% and 89.2% respectively. There were significant differences between the former two concentration groups and the latter two concentration groups ( P <0.005). Besides, there were also significant differences between 4 experiment groups and control group ( P <0.05). The result was coincident with that of microscopy observation and 3 H TdR marking experiment.
