摘 要:目的优选广痛消泡沫气雾剂中芍药苷的提取工艺。方法以芍药苷的含量为指标,采用L9(34)正交试验设计,对提取溶剂倍数(A)、提取次数(B)和提取时间(C)3个因素(每个因素设3个水平)进行考察,以确定最佳提取工艺。采用高效液相色谱法进行含量测定,Agilent extend色谱柱(4.6 mm×150 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(12∶88),流速为0.8 mL/min,柱温30℃,检测波长为230 nm。结果各因素对芍药苷提取量的影响大小顺序为A>B>C,最佳提取工艺为A3B2C1即加8倍水体积,提取2次,每次1h。验证试验结果显示,芍药苷的提取重量为449.5025 mg,提取量最多。结论该工艺合理,且稳定可行,为以后的生产和质量控制奠定了基础。Objective To optimize the extraction technique of paeoniflorin in Guangtongxiao Foam Aerosol.Methods Taking the content of paeoniflorin as index,the multiple of extraction solvent(A),number of extraction(B) and period of extraction(C) were observed by using L9(34) orthogonal design test for determining the best extraction technique.The HPLC procedure was performed on the chromatographic column of Agilent Extend(4.6 mm×150 mm,5 μm) at 30℃ and the mobile phase was acetonitrile-0.1% phosphoric acid solution in gradient elution program(12:88).The flow velocity was 0.8 mL/min and the detection wavelength was 230 nm.Results The sequence of influence of all factors on paeoniflorin extraction volume was ABC and the best extraction technique was A3B2C1(adding water by 8 times,extracting for 2 times and for 1 hour each time).The extractive weight of paeoniflorin was 449.502 2 mg,which was the most extraction quantity.Conclusion The technique is reasonable,stable and available,and provides a foundation for the further production and quality control of Guangtongxiao Foam Aerosol.
