摘 要:目的探讨人参皂苷Rg1抗同型半胱氨酸(Hcy)诱导人脐静脉内皮细胞(HUVECs)凋亡的作用及相关机制。方法体外培养HUVECs细胞,采用琼脂糖凝胶电泳和末端脱氧核糖核苷酸转移酶介导的缺口末端标记法(TUNEL)染色检测细胞凋亡情况,逆转录-聚合酶链反应(RT-PCR)和Western blotting分析各组内皮型一氧化氮合酶(eNOS)mRNA和蛋白质的表达。结果琼脂糖凝胶电泳显示:与对照组和Rg1组比较,Hcy组可见特征性DNA的"梯状"条带。TUNEL染色显示:与对照组和Rg1组比较Hcy组细胞凋亡率增加差异有统计学意义(P<0.01);RT-PCR显示:与对照组和Rg1组相比,Hcy组eNOS mRNA水平表达降低差异有统计学意义(P<0.01);与Hcy组相比,Hcy+Rg1组eNOS mRNA水平表达增加差异有统计学意义(P<0.01),但仍低于对照组和Rg1组,差异有统计学意义(P<0.01)。Western blotting显示:与对照组和Rg1组相比,Hcy组eNOS蛋白水平表达降低差异有统计学意义(P<0.01);与Hcy组相比,Hcy+Rg1组eNOS蛋白水平表达增加差异有统计学意义(P<0.01),但仍低于对照组和Rg1组,差异有统计学意义(P<0.01)。结论人参皂苷Rg1能够抑制Hcy诱导的人脐静脉内皮细胞凋亡,此作用可能与上调血管内皮细胞eNOS水平有关。Objective To investigate the inhibiting effect of ginsenoside Rg1 on apoptosis of human umbilical vein endothelial cells(HUVECs)induced by homocysteine(Hcy)and its possible mechanism.Methods HUVECs were incubated in vitro.The agarose gel electrophoresis and the terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling(TUNEL)were applied for the detection of apoptosis of HUVECs treated with Hcy and Rg1.RT-PCR and Western blotting were performed to determine the expression of endothelial nitric oxide synthase(eNOS).Results The results from the agarose gel electrophoresis displayed that DNA ladder was seen in HUVECs treated with Hcy group.The results from the TUNEL staining displayed that compared to the control group and Rg1 group,the number of apoptotic cells in Hcy group increased significantly(P0.01);compared to the Hcy group,the number of apoptotic cells in the Hcy+Rg1 group decreased markedly(P0.01),and the apoptotic percentage decreased from 33.733% to 9.200%,but the percentage was still higher than the control group and Rg1 group(P0.01).The results from the RT-PCR and Western blotting showed that compared to the control group and Rg1 group,the expression of eNOS decreased apparently in the Hcy group(P0.01);compared to the Hcy group,the expression of eNOS increased visibly in the Hcy+Rg1 group(P0.01),but the expression of eNOS was still lower than the control group and Rg1 group(P0.01).Conclusion Hcy can induce the apoptosis of HUVECs.Gisenoside Rg1 can protect HUVECs from the injury induced by Hcy.Upregulation of eNOS expression may be one of the possible mechanisms.
