摘 要:采用白血病祖细胞集落形成(CFUAML) 培养法,观察不同浓度人参总皂甙(TSPG) 对HL60、Meg01 白血病细胞株和急性髓细胞白血病患者的骨髓细胞集落形成的影响,并以正常粒单祖细胞(CFUGM)作比较。结果:正常和白血病祖细胞均对低浓度(5~25ug/ml) 的TSPG有反应,集落数提高26.1~35.4% ;中等浓度(75ug/ml) 的TSPG仍然刺激正常CFUGM 集落增加,但抑制HL60、Meg01 和CFUAML集落生长,抑制率分别为50.8% 、25 .1 % 和19.0% ;较高浓度(100ug/ml) 的TSPG对白血病祖细胞的抑制作用更明显,HL60 不能形成集落,对Meg01 和CFUAML的抑制率分别为53.3% 和28 .9 % ,而正常CFUGM 仍生长良好。提示TSPG对白血病祖细胞的影响不同于正常祖细胞。Colony forming culture method of leukemic ancestral cells was used to observe influence of total saponin of ginseng(TSPG)in different concentration to colony formation of HL-60.Meg-01 leukemic clone and marrow cell(CFU-AML)in the patients with acute myelocytic leukemia and also compared with normal granular ancestral cells(CFU-GM).The results showed that both normal and leukemic ancestral cells reacted to TSPG in the lower concentration(5~25ug/ml)and colony unit enhanced to 26.1~35.4%.TSPG in the medium concentration(75ug/ml)still stimulated increase in normal CFU-GM colony but inhibited colony growth of HL-60,Meg-01 and CFU-AMI.The inhibitory rates were respectively 50.8%,25.1% and 19.0%.The inhibitory function of TSPG in the higher concentration(100ug/ml)to leukemic ancestral cells was more obvious and its inhibitory rates to Meg-01 and CFU-AML were respectively 53.3% and 28.9%.But,normal CFU-GM still grew quite well.These results indicate that influence of TSPG to leukemic ancestral cell is different from its influence to normal ancestral cells and TSPG in different concentration shows either the proliferative or inhibitory functions.
