摘  要:从污泥中分离并筛选出一株具有较高絮凝活性的微生物絮凝剂产生菌,命名为Be-3。为了取得絮凝剂的最大活性并使其得到充分利用,对其培养条件进行了单因素优化,考察了微生物絮凝剂产生菌的特征和性质,实验结果表明,该菌产生絮凝剂的最佳培养条件为:碳源10 g.L-1乳糖和10 g.L-1蔗糖的混合物,氮源0.5 g.L-1尿素、0.2g.L-1酵母膏和0.5 g.L-1硝酸钾的混合物,无机盐为2 g.L-1磷酸二氢钾、5 g.L-1磷酸氢二钾和0.1 g.L-1氯化钠的混合物,培养基初始pH为6.5,培养温度为30℃,在摇床转速为120-140 r.min-1的培养箱中培养42 h时,絮凝活性可达98.04%。通过生理生化实验分析,该絮凝剂产生菌为革兰氏阳性菌,主要成分为多糖。用Be-3所产絮凝剂处理不同废水后,废水中的CODcr、NH3-N和浊度的去除率都比较高,说明该絮凝剂在废水处理中具有很好的应用前景。The strain of high-efficiency bioflocculant-producting bacteria,which was named Be-3,was screened from the sludge,in order to get the highest flocculating activities and make full use of the flocculent,their characteristics and optimal culturing condition were studied in the paper.The results of the experiment showed that the optimal culturing condition was as follows: the carbon source was the composition of 10g·L-1 cane sugar and 10 g·L-1 lactose,the nitrogen source was the composition of 0.5 g·L-1 urea,0.2 g·L-1 yeast extract and 0.5 g·L-1 potassium nitrate,the inorganic salt was the composition of 2 g·L-1 potassium dihydrogen phosphate,5 g·L-1 potassium phosphate dibasic and 0.1 g·L-1 sodium chloride.When the Be-3 was cultured at 30 ℃ with the initial pH being 6.5 and the rotation speed-140 r·min-1 for 42 h,the flocculating activities achieved 98.04%,the strain was gram-positive,and the main components of flocculants was polysaccharide in physiological biochemical test.When the strain was applied to wastewater treatment,the removal rate of CODcr,NH3-N and turbidity were comparatively good.The results suggested that the strain had good application prospect in practice.
