PCR-SSP在异基因造血干细胞移植供体选择中的应用

摘　　要：目的 :建立常规的 PCR- SSP基因分型方法。方法 :对 16名患者及其家系 HL A- DR进行血清学和 PCR- SSP分型。结果 :血清学分型 7对供、受体相合 ,而 PCR- SSP基因分型只有其中 5对相合 ,另外 2对 1对为错判、1对为 DR2两个亚型 DR15、DR16间的错判。结论 :PCR- SSP基因分型是一种快速、准确和可行的 HLA分型方法 ,具有广阔的临床应用前景。Objective:To build frequent method of HLA DNA typing by PCR-SSP.Methods:16 patients and their families were done for HLA-DR typing by serology and PCR-SSP respectively.Results:The serology HLA-typig showed that 7 pairs of donors and receipts were HLA-matched,however,PCR-SSP HLA-typing showed that 5 pairs of donors and receipts are HLA-matched,among other 2 pairs,one was wrong HLA-typing,one was wrong HLA-typing between DR15 and DR16 subtype of DR2 by serology.Conclusions:PCR-SSP is a rapid,precise and reliablc HLA-typing mothod,it have very wide prospect in clinical HLA-typing.
