摘  要:目的研究低强度电磁辐射对原代培养的小鼠脑皮质神经元的损伤效应,为制定防护电磁辐射对中枢神经系统危害的措施提供依据。方法将原代培养的小鼠脑皮质神经元给予900 MHz平均功率密度为8 mW/cm2的电磁波持续辐射1 h、6 h、12 h,辐射后分别用CCK-8试剂盒检测细胞活力,用Annexin V-FITC/PI双标记试剂盒流式细胞仪检测细胞凋亡率。结果细胞活力分析显示,与对照组细胞比较,持续辐射1 h组差异不显著;辐射6 h组细胞活力则降低22%(P<0.05),辐射12 h组细胞活力则降低37%(P<0.05);Annexin V-FITC/PI细胞凋亡分析显示,与对照组细胞凋亡率(5.6%)比较,持续辐射1 h组差异不显著,辐射6 h组细胞凋亡率达17.6%(P<0.05),辐射12 h组达30.8%(P<0.05)。结论低强度非致热效应电磁波持续辐射时间长于6 h后可引起原代培养的小鼠脑皮质神经元损伤,并产生凋亡。Objective To investigate the effects of exposure to low-intensity electromagnetic fields on cerebral cortical neurons in mice,so as to provide the basis for working out protective measures against electromagnetic radiation damage.Methods Primary neonatal mouse neurons were exposed to 900 MHz electromagnetic fields with power density of 8 mW/cm2 for 1 h,6 h,and 12 h respectively.Analysis of cell viability was performed by CCK-8 kit,and apoptosis was detected by Annexin V-fluorescein isothiocyanate(FITC)/propidium(PI) kit and flow cytometry after exposure.Results The cell viability of neurons exposed to power density of 8 mW/cm2 for 1 h was generally not affected,and the measurement of apoptosis showed no differences between exposed neurons and unexposed control neurons too.However,significant changes were observed in the neurons exposed to electromagnetic fields for 6 h or 12 h.The cell viability in neurons exposed for 6 h or 12 h decreased 22% and 37% more respectively than in the unexposed control neurons.The cell apoptosis rate was 5.6% in control,but in exposed ones for 6 h,or 12 h increased to 17.6% or 30.8% respectively(P0.05).Conclusion Exposure to low-intensity electromagnetic fields for more than 6 h might cause primary cerebral cortical neurons damage and apoptosis.
