EFFECT OF HEAT STRESS ON SPATIAL MEMORY AND THE CHANGES OF HIPPOCAMAPAL STRUCTURE IN MICE

摘  要:目的研究热应激环境下小鼠空间工作记忆、空间参考记忆的变化,同时观察海马对超微结构的影响,为研究热损伤机制及防护措施提供依据。方法昆明种小鼠30只,随机分为热应激组(40℃,60 min)、对照组和电镜观察组。采用恒温恒湿动物箱建立热应激模型,利用Morris水迷宫观察小鼠空间工作记忆、参考记忆情况,透射电镜观察小鼠海马组织的细胞形态结构。结果热应激组的空间参考记忆和空间工作记忆时间分别为(50.7±7.1)s和(46.2±6.8)s,明显长于对照组的空间参考记忆时间(41.0±8.2)s和空间工作记忆时间(37.7±7.4)s(P<0.05,P<0.01),但热应激组小鼠海马组织的形态结构完整。结论 40℃热应激60 min,小鼠的记忆能力下降,而海马超微结构未发生变化。Objective To study the changes of spatial reference memory and spatial working memory performance,and ultrastructure of hippocampal neurons in adult mice after acute heat stress,so as to provide the basis for studying the mechanism of heat injury,and working out protective measures against it.Methods Thirty mice were randomly divided into three groups,heat stress group,control group and electron microscope group.Adult mice were placed in insulated cages and exposed to 40 ℃,RH 60% for 60 min.Spatial reference memory and working memory performance were tested by Morris water maze.The hippocampal neurons were examined with transmission electron microscope. Results The spatial reference memory(50.7±7.1)s and working memory(46.2±6.8)s in heat stress group were longer than those in control group((41.0±8.2)s,(37.7±7.4)s respectively).But the neuronal ultrastructure did not change.Conclusion The memory ability of mice was decreased in heat stress at 40 ℃ for 60 min while the change of hippocampus ultrastructure was not observed.
