摘 要:测定了3个兰州鲇群体,即野生兰州鲇(WS)、兰州鲇亲本(QS)、人工繁育鲇子一代(ZS),共56尾样本的线粒体细胞色素b基因,运用MEGA 4.0、DNASP等软件对样本的968 bp线粒体Cytb基因序列进行了分析。结果显示:共检测到17种单倍型、99个变异位点,其中有93个是多态位点(包含74个简约信息位点),总变异率为10.23%;对同源序列碱基组成分析显示,T、C、A、G碱基的平均含量分别为26.87%、29.46%、29.16%、14.53%,A+T含量大于G+C含量;Kimura′s 2-parameter模型分析得到兰州鲇3个群体之间的遗传距离介于0.001 1~0.084 4之间,兰州鲇3个群体(QS、ZS、WS)单倍型多样性指数分别是(0.916±0.027)、(0.714±0.071)、(0.857±0.039),核苷酸多样性指数分别(0.008 01±0.000 006 9)、(0.002 87±0.000 001 2)、(0.019 55±0.000 025 0).In this study,the mitochondrial DNA cytochrome b of 56 samples representing three groups of Silurus lanzhouensis were sequenced.The MEGA 4.0 and DNASP software were used to analyze the 968 bp gene fragment.The results showed that:17 haplotypes,99 sites of mutation were detected accounting for 10.23 % of total sequence,93 sites were polymorphism sites,in which 74 sites were parsimony informative sites.After being aligned,the average basic compositions of all sequences were as follows:26.87 % A,29.46 % C,29.16 % T and 14.53 % G,while the average basic compositions of A+T were bigger than G+C.Genetic distance were calculated by the Kimura′s 2-parameter model,which ranged from 0.001 1 to 0.084 4,haplotype diversity of three groups of Silurus Lanzhouensis(QS,ZS and WS) were(0.916±0.027),(0.714±0.0710) and(0.857±0.039),respectively,and nucleotide diversity of those three groups were(0.008 01±0.000 006 9),(0.002 87±0.000 001 2) and(0.019 55±0.000 025 0),respectively.
