摘 要:为了解鸡传染性贫血病毒(CIAV)广西分离株全基因变异情况,设计3对特异性引物,对CIAV广西分离株(GXC060821)的全基因进行PCR扩增、克隆和序列分析。结果显示,广西分离株的全基因为2 292个核苷酸,含有3个互相重叠的开放阅读框(ORF)。序列分析表明,本CIAV分离株与国内外其他31个CIAV毒株比较核苷酸的同源性在96.1%~98.5%之间,推导氨基酸的同源性在89.8%~94.2%之间。全基因系统发育进化树分析显示,广西分离株和中国的TJBD40株、LF4株都处于一个分支,它们的亲缘关系较近,而与美国的98D06073分离株及中国的HA4分离株的亲缘关系较远。In order to understand the variation tendency of Chicken infectious anemia virus(CIAV) wild strains isolated from Guangxi,one pair of special primers were designed and synthesized according to complete gene sequences of CIAV strains in GenBank.The complete genome of CIAV isolates from Guangxi were amplified by polymerase chain reaction(PCR).The full-length complete genome of eleven CIAV isolates in Guangxi were sequenced after the products of amplification being cloned.All complete nucleotide sequences of CIAV isolates in Guangxi are 2 292 bp long,it contained 3 overlapped ORF.The results of complete gene analyses indicated that comparison of complete sequences and the deduced amino acid sequences of CIAV isolates with 31 other published CIAV reveals the homologies of the nucleotide between 96.1%-98.5% and the homologies of the deduced amino acid sequences are between 89.8%-94.2%.Phylogenetic tree analysis of entire genome indicated that the isolates of Guangxi were close to the strain TJBD40 and LF4 from China,and had a distant with USA isolate 98D06073 and China HA4 isolate.
