摘 要:为了解耐药质粒消除对大肠埃希菌耐药性及其他生理特性的影响,参考前期试验分离出的8株携带aac(6’)-Ib质粒耐药基因的大肠埃希菌的药敏试验。选取1株耐药性最强菌株用高温及SDS进行耐药质粒消除,PCR检测aac(6’)-Ib基因。结果显示,耐药质粒成功消除,耐药性消失,且细菌在不含抗生素条件下培养,耐药性消除状态可稳定遗传;但在含卡那霉素条件下连续传代,其耐药性可迅速恢复,第4代即可产生耐药性。体外生长曲线显示,耐药质粒消除株生长较快,且较早进入对数期。以上数据表明大肠埃希菌多重耐药性主要由质粒决定,研究开发新型耐药质粒消除剂对克服大肠埃希菌耐药性具有重要意义。To understand the drug resistance and the other physiological property after plasmid elimination treatment of E.coli.The E.coli with plasmid containing aac(6')-Ib gene was used for drug sensitivity test.The drug sensitivity showed that the strains almost resisted all of the 8 antibiotics including Aminoglycosides and Quinolones.In this study one strain of highest drug-fast was reversed by high temperature and SDS.PCR results showed that the resistant gene named aac(6')-Ib was eliminated,and the drug resistance was disappeared.When the strain was cultured without antibiotic,the drug sensitivity was constant.But the environment of kanamycin could recover the drug resistance after 4 generations.The strain without drug-resistance plasmid growed faster,and went into logarithmic phase earlier.Therefore,drug resistance of E.coli caused by plasmid.The strategy to eliminate the plasmids would be an effective way to deal with the multiresistance issue.
