摘 要:分离发病兔场皮肤真菌病的病原,并对其内转录间隔区(ITS)序列进行分析鉴定。采集病兔患病部位的皮屑、痂皮进行培养,并根据形态学的特点进行鉴定;然后采用真菌内转录间隔区通用引物对该区域进行PCR扩增测序,并将该区域序列与GenBank中的核酸数据库进行比对分析,确定病原真菌的生物学分类,最后对其亲缘关系进行系统发育分析。分离得到的病原真菌经形态学初步鉴定为小孢子菌属;序列比对和系统发育分析表明,该菌与Microsporum gypseumstrain WCH-MG001(序列号:GU348990.1)的内转录间隔区(ITS)序列具有99%的同源性,在系统发育树上属于同一分支,从而确定该菌为石膏样小孢子菌。Rabbit dermatophytes were isolated and their internal transcribed spacers(ITS) sequences were analyzed and identified.The collected scurf and scab were cultured,and then identified by morphological traits;the fungal ITS region-specific primers were used to perform amplification and sequencing of this region.Through the blast analysis between ITS sequences and GenBank nucleic acid database,the biological classifications were identified and finally their relationships were analyzed according to the phylogenetic tree.The isolated rabbit dermatophytes were preliminary identified as Microsporum by morphological characteristics;the results of sequence analysis and phylogenetic tree showed that the isolated fungi and Microsporum gypseum strain WCH-MG001(Accession Number:GU348990.1)belonged to one group with 99% homology,indicating that the isolated fungi were Microsporum gypseum.
