摘 要:目的探讨人骨髓间充质干细胞(hBMSCs)体外分离培养、鉴定条件及方法,探讨其作为种子细胞构建血管化组织工程皮肤可行性。方法用密度梯度离心法分离骨髓间充质干细胞,筛选贴壁细胞培养并传代。观察细胞形态,绘制细胞生长曲线,检验细胞CD44、CD29、CD14、CD45等表面标记。采用定向诱导分化方法体外鉴定hBMSCs所具有的多项分化能力。结果骨髓密度梯度离心法分离hBMSCs,接种培养瓶后呈克隆样贴壁生长。在特定培养基中hBMSCs可向成骨、成脂及成软骨方向转化。结论成功建立hBMSCs分离培养鉴定体系。所培养的hBMSCs可以成为皮肤组织工程较为理想的种子细胞。Objective To investigate the identification and culture of isolated human bone marrow mesenchymal stem cells ( hBMSC), and to study the feasibility of construction of vascularized tissue - engineered skin using hMSCs as seed cells. Methods Human BMSCs were isolated and cultured for passage using density gradient centrifugation and ad- herent cells filter method, respectively. Cell growth curve was drawn according to morphologie observation. Meanwhile, surface markers, including CD44, CD29, CD14, CIM5 and etc, were detected. Pluripotent hBMSCs were indentified with directed induced differentiation in vitro. Results Isolated hBMSCs presented adherent clone - like growth after inocula- tion in culture bottle. Osteogenie, adipogenic or chondrogenic differentiations were revealed in hBMSCs cultured in specific medium. Conclusion Human BMSCs are successfully isolated, cultured and identified, and can be used as seed cells of the skin tissue engineering.
