摘 要:目的初步探讨单核细胞趋化蛋白-1(MCP-1)﹑Ⅳ型胶原蛋白(Ⅳcollagen)和血清在单核细胞迁移过程中的作用。方法体外单核细胞迁移模型的建立,在体积分数10%FBS/1μgⅣcollagen和体积分数0%FBS/1μgⅣcollagen的实验条件下添加650 ng/mL MCP-1与0 ng/mL MCP-1。流式细胞术检测迁移前后单核细胞表面抗原CD14阳性率。结果在体积分数0%FBS/1μgⅣcollagen的实验条件加入650 ng/mL MCP-1时细胞生长良好。迁移前CD14细胞阳性率19.6%,迁移后CD14细胞阳性率1.5%。结论单核细胞在FBS体积分数为0%,Ⅳcollagen浓度为1μg、MCP-1浓度为650 ng/mL实验条件时细胞迁移数量较多。Objective To investigate the effects of MCP - 1, collagen IV and serum on monocyte migration. Methods To establish the monocyte migration model in vitro. MCP - 1 at the concentration of 650 ng/mL or 0 ng/mL was added into FBS in volume fraction 10% and 0% with 1 μg collagen IV for stimulation. CD14 on monocytes was detec- ted by flow cytometry before and after migration. Results CD14 positive rates of monocytes were 19.6% and 1.5% be- fore and after migration, respectively. Significantly better growth was observed on monocytes with I p.g collagen Ⅳ at 650 ng/mL of MCP - 1 and FBS in 0% volume fraction. Conclusion Monocytes in 1 μg collagen IV at 650 ng/mL MCP - 1 with FBS in 0% volume fraction present maximal migration ability.
