枇杷茎尖愈伤组织诱导及种质体外保存研究

摘　　要：以国家果树种质资源枇杷圃中12份枇杷种质为试材,开展了枇杷茎尖愈伤组织的诱导和种质体外保存研究。结果表明,将春季采集的枇杷茎尖不经水冲洗,直接接种于MS+6-BA 1.0 mg.L-1+NAA 1.0mg.L-1培养基上培养,较容易诱导出愈伤组织;不同基因型的枇杷茎尖诱导出的愈伤组织存在较大差异,诱导率最高的是野生92号,为96.2%;诱导率最低的是黄岩5号,没有诱导出愈伤组织。采用MS+NAA3.0 mg.L-1+6-BA1.0 mg.L-1+蔗糖6%(2～3代)→MS培养基(1～2代,其中有一次使用液体培养)→MS+NAA3.0 mg.L-1+6-BA1.0 mg.L-1+蔗糖6%(2～3代)循环继代路线,在弱光(光照度150 lx)下培养,能较好地保存枇杷茎尖愈伤组织,胚性没有减弱,生长势较好且仍为黄白色颗粒状。试验结果为枇杷种质体外保存、原生质体培养、细胞融合、遗传转化及离体诱变等研究打下基础。12 loquat shoot tips,collected from National Field Genebank for Loquat in Fuzhou,were cultured to induce callus and preserved in vitro.The results showed that the shoot tips in spring without washing was more easily to induce callus if inoculated directly to MS supplemented with 1.0 mg·L-1 6-BA and 1.0 mg·L-1 NAA.The inducing ability for callus in different genotypic loquat shoot tips was different.The highest induction rate was wide loquat-92（96.2%）.The lowest induction rate was Huangyan-5 which couldnt induce callus.The callus were conserved better under weak light（illuminance 150 lx） with a cycle subculture process： MS supplemented with 3.0 mg·L-1 NAA and 1.0 mg·L-1 6-BA and 6% sucrose（2-3 generation） →MS（1-2 generation,with once liquid culture） → MS supplemented with 3.0 mg·L-1 NAA and 1.0 mg·L-1 6-BA and 6% sucrose（2-3 generation）.At this condition the callus grew preferably and was yellow-white granular.The results will lay the foundation for loquat germplasm preservation,protoplast culture,cell fusion,genetic transformation and mutagenesis in vitro.
