摘　　要：目的研究rPVL对Balb/C小鼠肺组织损伤及肺组织匀浆中细胞因子的影响。方法 20只成年Balb/C小鼠分两组,对照组、rPVL组。对照组注射生理盐水,rPVL组注射rPVL,两组分别于注射后3 h、9 h处死小鼠,每个时间段5只,ELISA方法测肺组织匀浆中TNF-α、IL-8水平,RT-PCR方法测肺组织匀浆TNF-α、IL-8mRNA的表达。结果 rPVL刺激小鼠后3 h、9 h肺组织匀浆TNF-α、IL-8蛋白含量、mRNA表达量均高于正常对照组,差异有统计学意义(P<0.05),rPVL组TNF-α、IL-8蛋白含量、mRNA表达量随时间延长而升高,差异有统计学意义(P<0.05)。结论 rPVL能引起小鼠肺组织严重损伤,且肺组织匀浆中TNF-α、IL-8大量释放,提示细胞因子在rPVL对小鼠肺组织损伤过程中起重要作用。Objective To investigate the effect of recombinant panton-valentine leukocidin（rPVL） on the changes of TNF-α、IL-8 in Balb/C mice lung tissue.Methods A total of 20 adult Balb/C mice were divided into normal group,rPVLgroup.The normal group was injected with NS,the rPVL group was injected with rPVL.The mice were killed after injection 3 h,9 h respectively.The content of TNF-α,IL-8 and the expression of TNF-α,IL-8 mRNA were determined by ELISA or RT-PCR respectively.Results The TNF-α,IL-8 content and the expression of TNF-α,IL-8 mRNA in rPVL group were higher than the normal control group at 3 h、9 h after rPVL stimulating the mouse lung tissue（P〈0.05）,The TNF-α,IL-8 content and the expression of TNF-α,IL-8 mRNA in rPVL group were also increased with time（P〈0.05）.Conclusion A large number of TNF-α,IL-8 were released when rPVL stimulated mice lung tissue,suggesting that cytokines play an important role in rPVL-stimulated mice lung tissue.
