泻心汤配方颗粒和传统汤剂中盐酸小檗碱含量及体外抗菌作用比较

摘　　要：目的考察泻心汤传统汤剂和配方颗粒汤剂中盐酸小檗碱的含量变化,并对泻心汤传统汤剂和配方颗粒汤剂进行体外抗菌作用比较。方法建立高效液相色谱方法,Diamonsil-C18色谱柱(4.6 mm×250 mm,5μm),流动相:乙腈-0.05mol.L-1NaH2PO4磷酸调pH值为3(30∶70),流速:1m.lmin-1,柱温:30℃,检测波长为350nm处测定盐酸小檗碱。结果 (1)泻心汤配方颗粒汤剂中盐酸小檗碱的平均含量显著高于传统汤剂汤剂的平均含量;(2)泻心汤传统汤剂对黑曲霉菌的抑菌作用大于配方颗粒汤剂、配方颗粒汤剂对大肠杆菌的抑菌作用大于传统汤剂;合煎液传统汤剂和配方颗粒汤剂对金黄色葡萄球菌、白色葡萄球菌、乙型溶血性链球菌都有抑制作用。结论与传统汤剂比较,配方颗粒汤剂中盐酸小檗碱的含量较高,对于黑曲霉菌传统汤剂抑菌作用更强,而对于大肠杆菌配方颗粒汤剂抑菌作用更强。Aim To compare the content of berberine hydrochloride in hejian decoction（the traditional Chinese herbs mixed together then boiled with water）of XieXin Tang and the fenjian decoction（the single traditional Chinese herb boiled with water separately,before the Abstracts mixed）of XieXin Tang（composed of Scutellaria baicalensis,Rheum palmatum and Coptis chinensis）and to compare their anti-bacterial activities in vitro.Methods A HPLC method was established with a Diamonsil-C18 column（4.6 mm× 250 mm,5 μm）,a mobile phase of acetonitrile-0.05 mol·L-1sodium dihydrogen phosphate solution（adjust pH to 3 with phosphoric acid）（30∶ 70）,a flow rate of 1 mL·min-1 and a detection wavelength of 350 nm in order to determine the contents of berberine hydrochloride.Minimal bacterial inhibitory concentration（MIC）and minimal bactericidal concentration（MBC）antagonizing the common bacteria in different decoctions were measured in vitro by dilution method.Results The average content of berberine hydrochloride of the fenjian decoction was higher than that of the hejian decoction.The hejian decoctions could display the inhibitory bactericidal activity to A.niger was stronger than that of the fenjian decoction.And to proteus,the inhibitory bactericidal activity of the fenjian decoction was stronger than that of the hejian decoction.The hejian decoctions and the hejian decoctions could display the inhibitory bactericidal activity to Staphylococcus aureus,S.albus,β-hemolyticstreptococcus.Conclusion Compared with that of the hejian decoction,the content of berberine hydrochloride of the fenjian decoction was higher,to A.niger,the inhibitory bactericidal activity of the hejian decoctions was stronger than the fenjian decoction,but to colon bacillus,the inhibitory bactericidal activity of the fenjian decoctions was stronger than the hejian decoction.
