摘 要:目的比较两种免疫程序和途径制备抗血清的效价,并在免疫学溶血反应中的运用效果比较,寻求获取符合实验教学需要的高效价溶血素的较佳方法。方法采用不同浓度的绵羊红细胞(Sheep red blood cells,SRBC)间隔1天和间隔3天腹腔、耳静脉免疫家兔,用血球凝集法浓度为1.5%SRBC悬液检测并比较两种免疫程序和途径所制备溶血素的效价结果免疫后获得的溶血素效价几何平均滴度,A组脱纤维绵羊RBC全血皮内多点注射家兔为1:22286.09,B组10%脱纤维绵羊RBC悬液耳静脉注射家兔为1:72407.73。结论 10%脱纤维SRBC悬液耳静脉注射家兔制备的溶血素效价高于脱纤维SRBC全血皮内多点免疫家兔,是获取实验教学用溶血素的较佳免疫程序。Objective To compare two types of anti-Sheep RBC hemolysim antiserum titers processed by different immunizations and select the better procedure to obtain high titer antiserums for experimental teaching.Methods 10 rabbits were immunized with defibrinated Sheep red blood cells(SRBC) intraperitoneally every 2 days(Group A).Meanwhile,another 10 rabbits were immunized with defibrinated SRBC intravenously every 4 days(Group B).The antiserum titers were detected by Hemagglutination inhibition test.Results The geometric mean of anti-SRBC antiserum titers in Group A and B were 1:22286.09 and 1:72407.73 respectively,compared to unlike immunization group.Conclusion The higher antiserum titers were obtained through 3 day-interval intravenous immunization than 1 day-interval intraperitoneal immunization.The procedure of 3 day-interval intravenous immunization with 10% defibrinated SRBC is the better one for hemolysin preparation of experimental teaching
