摘 要:该试验从亚硝化和全程硝化两种不同硝化类型的好氧流化床反应器的固定化包埋颗粒中提取细菌总基因组DNA,采用聚合酶链式反应-变性梯度凝胶电泳技术(PCR-DGGE)对系统中的氨氧化菌(ammonia-oxidizing bacteria,AOB)和亚硝酸盐氧化菌(nitrite-oxidizing bacteria,NOB)群落结构进行了解析,根据序列数据进行同源性分析并建立系统发育树。结果表明:经过较短的驯化时间后,固定化颗粒反应器可形成优势菌,改变运行条件或经过长时间搁置后,颗粒中生态群落结构保持稳定;反应器中AOB较单一,以Nitrosomonas sp.为主,但具备一定抗冲击负荷能力,NOB以Nitrobacter sp.为主,全程硝化反应器中具有亚硝化反应器中所不具备的Nitrobacter sp.219、Nitrobacter sp.263。Total bacterial genomic DNA was extracted from the immobilized pellets in partial nitrification and nitrification aerobic fluidized bed reactors,respectively.The microbial community structures were studied by polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE).The sequences were used for homology analysis and phylogenetic trees construction.The results indicate that the immobilized pellets reactor could achieve stabilization after a short period of acclimation and no significant distinction was observed between the dominant bacteria under different operation conditions or even after a long time of placement.Nitrosomonas sp.is the most predominant ammonium oxidizing bacteria(AOB) in the system with a capability against the shock loading and Nitrobacter sp.constituted the most part of nitrite oxidizing bacteria(NOB) in the system.Nitrobacter sp.219 and Nitrobacter sp.263,which play a key role in the nitrification system,are absent in the partial nitrification system.
