紫苏叶挥发油化学成分分析及其抗炎机制研究

摘　　要：目的明确紫苏叶挥发油化学成分组成及其抗炎作用机制。方法采用GC-MS对水蒸气蒸馏提取所得的紫苏叶挥发油进行组成成分分析。采用角叉菜胶致去肾上腺大鼠足跖肿胀模型,角叉菜胶致大鼠胸膜炎模型,观察紫苏叶挥发油对不同炎症模型的抗炎作用,测定大鼠足跖浸出液中前列腺素E2(PGE2)含量,大鼠胸腔渗出液中蛋白、白细胞数(WBC)、NO含量;同时,测定正常小鼠肾上腺重量及肾上腺中Vit C的含量。结果从紫苏叶挥发油中共鉴定出32种成分,其主要含有柠檬烯(18.3%)、紫苏醛(16.5%)、紫苏酮(12.4%)、芹菜脑(9.3%)和反式丁香烯(12.0%)。紫苏叶挥发油能显著减轻角叉菜胶致去肾上腺大鼠足跖肿胀,降低炎症组织PGE2含量,抑制胸腔渗出液中蛋白含量和白细胞数,降低NO含量。但不影响正常小鼠的肾上腺重量及肾上腺中Vit C的含量。结论紫苏叶挥发油具有明显的抗炎作用,其抗炎作用可能与其抑制白细胞游走、减少蛋白质渗出、清除氧自由基、抑制PGE2等炎症介质生成有关,而与肾上腺皮质系统无关。OBJECTIVE To investigate the chemical composition and anti-inflammatory activities of essential oil hydrodistilled from Perill frutescens leaf（OPF）.METHODS The essential oils of Perill frutescens leaf were analyzed by GC-MS.Inflammation induced by carrageenan in the hind paw of bilaterally adrenalectomize Wistar rat and in pleurisy of Wistar rat were applied for anti-inflammatory experiments.Then the content of protein,the amount of white blood cell（WBC）,prostaglandin E2（PGE2）and NO were determined.The weight of adrenal gland and the content of Vitamin C in adrenal gland of normal mice were measured as well.RESULTS Thirty two compounds was identified from OPF,of which Limonene（18.3%）,perillaketone（12.4%）,l-perillaldehyde（16.5%）,apiole（9.3%）,and trans-caryophyllene（12.0%）were the major compounds.Total flavonoids could inhibit vas opermeability and engorgement of inflammatory texture,decrease total protein content in efflusion and the amount of white blood cell,reduce PGE2 and NO contents.However,it had no effects on the weight of adrenal and the content of Vitamin C in normal mice.CONCLUSION The mechanisms of anti-inflammatory action of OPF are related to the reduction of the migration of neutrophils and exudation of protein,scavenging capacity of oxygen free radical,reduction of PGE2 but not depended on the adrenal gland.
