摘 要:探讨了采用苏丹黑B染料染色处理微藻细胞技术估测微藻油脂含量的可行性.结果表明,微藻细胞经苏丹黑B染色处理后在645 nm光波长处有最大吸收峰值A645,微藻油脂含量与经苏丹黑B染色处理后微藻细胞的吸光值A645呈明显的相关性.建立了考察的7种微藻油脂含量与吸光值A645的线性回归方程,线性关系显著(p〈0.01).基于藻湿重,微藻油脂含量的线性范围为1.79~39.63 mg/g,估算的相对偏差〈10%.因而,采用苏丹黑B染色处理微藻细胞技术适宜用来检测微藻的油脂含量.With continually rising fuel prices,biofuel as an alternative fuel is attracting more and more attention.Because of the high lipid content and the rapid increase in biomass,microalgae that grow in aquatic environments and use light and carbon dioxide to create biomass have been recognized as potentially good material sources for biofuel production in the future.Utilization of the microalgal lipid to produce biofuel has a promising future.Rapid and effective detection of the microalgal lipid content is important in scanning microalgal samples and determining suitable species for biofuel production.This study investigates the feasibility of assessing the microalgal lipid content by staining microalgal cells with the dye Sudan black B.The results show that microalgal cells stained with Sudan black B have maximum absorbance(A645) at a wavelength of 645 nm.The content of cell lipid(y) is related to the peak absorbance A645(x) of microalgal cells and can be expressed with a regression equation of the form y = Kx + b.Linear regression equations for cell lipid content and absorbance A645 were obtained and demonstrated similar linear relationships for the seven species of microalgae we studied(Chlorella elliptica,Platymonas helgolandica Tsingtaoensis,Phaeodactylum tricornutum,Isochrysis galbana,Dicrateria inornata,Dunaliella salina and Chlamydomonas reinhardi,p 0.01).For different microalgal species,the regression coefficient K and the constant b respectively varied from 55.818 to 64.131 and-1.6787 to 0.5765,with determination coefficient R2 ranging from 0.985 4 to 0.999 7.The linear range was between 1.79 and 39.63 mg/g in detection the microalgal lipid content on the basis of wet weight with relative deviation less than 10%.The study indicated that it is suitable to estimate the lipid content of microalgal cells by staining them with Sudan black B.
