鸭卵巢组织基因差异表达与繁殖性状杂种优势率相关性分析

摘  要:采用mRNA差异显示技术检测了产蛋高峰期高邮鸭、金定鸭及正反交组合群体卵巢组织中的基因差异表达,并分析了差异表达模式与繁殖性状杂种优势率的相关性。研究结果表明:杂交组合在开产日龄、42周龄产蛋数性状上表现出明显的杂种优势,杂种优势率>±10%;16对引物组合共扩增出326条cDNA片段,能够重现的有248条,重现率为76.03%,发现7种差异表达模式;相关性分析显示开产日龄、42周龄产蛋数杂种优势率与杂种超强表达模式呈极显著正相关(P<0.01),与杂种特异表达呈极显著负相关(P<0.01);共显性表达模式与开产日龄杂种优势率呈显著正相关(P<0.05);7种差异表达模式与42周龄种蛋受精率和受精蛋孵化率杂种优势率2个指标之间并不存在显著相关性(P>0.05)。mRNA differential display method was used to detect the differential gene expression of ovary tissues between two parental breeds Gaoyou,Jinding duck and their hybrids at laying peak. The correlation between the differential gene expression types and heterosis percentage of reproduction traits were analyzed as well. The results showed that comparing to parental breeds,two hybrid populations had visible heterosis in age of the in first laying day and egg production of 42 weeks age,with more than ten heterosis percentage. Total of 248 recurrent differential eDNA fragments from 326 were displayed in the cross combinations with 16 pairs of primers,and seven differential expression patterns were found. Correlation analysis indicated that there were significantly positive correlation (P〈0.01) between the heterosis percentage of age of the first laying day and egg production of 42 weeks age and over-expression in hybrids,while had significantly negative correlation (P〈0.01) with hybrid-specific expressed pattern. Co-dominant expressed pattern had significantly positive correlation (P〈0.05) with heterosis percentage of age of the first laying day. However,no significant correlation were found between heterosis percentage of egg fertilizing rate and hatching rate of 42 weeks age and detected seven differential expression patterns (P〉0.05).
