芎芷地龙汤对偏头痛模型大鼠脑组织痛觉传导通路c-fos和5-HT蛋白表达的影响  被引量:15

摘  要:目的研究芎芷地龙汤对偏头痛模型大鼠的镇痛作用机制。方法将48只健康雄性SD大鼠随机为生理盐水组、模型组、舒马普坦组、中药组,每组12只。皮下注射硝酸甘油制备偏头痛动物模型,经药物干预后在4h、6h取材,测定三叉神经脊束核(SPN5)和中脑导水管周围灰质(PAG)中细胞癌基因fos(c-fos)和5-羟色胺(5-HT)的阳性细胞数。结果 SPN5中蛋白表达:中药组c-fos阳性细胞数减少,4h多于6h,与模型组比较差异有统计学意义(P<0.01);5-HT阳性细胞数增多,4h稍多于6h,与模型组比较有统计学差异(P<0.05或P<0.01)。PAG中蛋白表达:中药组c-fos阳性细胞数减少,4h多于6h,与模型组比较有统计学差异(P<0.01);中药组5-HT阳性细胞数增加更多,6h多于4h,与模型组比较有统计学差异(P<0.01)。结论芎芷地龙汤可以减少由硝酸甘油诱导的c-fos的表达,增加5-HT的表达,进一步减少SPN5神经元的兴奋性,激动5-HT下行镇痛系统,且可持续较长时间,增高痛阈,阻止痛觉信息的传递,起到镇痛作用。Objective To study the mechanism of analgesic effect of Xiongzhi Dilong Decoction [Decoction of Chuanxiong (Rhizoma Ligustici Chuangxiong),Baizhi (Radix Angelicae Dahuricae),Dilong (Lumbricus)] on the migraine model rats. Methods The 48 healthy male SD rats were randomized into 4 groups:normal saline group,model group,Sumatriptan group,and Chinese medicine group,with 12 in each. The migraine models were made by subcutanous injection of nitroglycerin,and the brain tissue samples were collected 4h and 6h after medication intervention. The immunohistochemical staining technique was then conducted to determine the expression of celluar oncogene fos (c-fos) and 5-hydroxytryptamine (5-HT) in SPN 5 and PAG. Results Expression in SPN 5:A reduction of c-fos expression in the Chinese medicine group was observed,with a decrease in expression from 4h to 6h. There was statistical difference in comparing with that of the model group (P0.01). An increase of 5-HT expression in the Chinese medicine group was noticed,with a slight decrease in expression from 4h to 6h. There was statistical difference in comparing with that of the model group (P0.05,P0.01). Expression in PAG:c-fos expression in the Chinese medicine group was reduced,with decrease in expression from 4h to 6h. There was statistical difference in comparing with that of the model group (P0.01). The increase of 5-HT expression in the Chinese medicine group was more significant,and the positive cell was more at 6h than at 4h. There was statistical difference in comparing with that of the model group (P0.01). Conclusion Xiongzhi Dilong Decoction is able to reduce the c-fos expression induced by nitroglycerin,increase the 5-HT expression,hence depressing excitability in neurons of SPN 5 by stimulating the descending 5-HT analgesic pathway. This effect can last for a long period of time,to make the pain threshold increased and the transmission of nociceptive information inhibited.
