摘  要:目的探讨中药复方金欣浓缩液保留灌肠治疗呼吸道合胞病毒(RSV)肺炎的作用机制。方法将56只SD大鼠随机分为正常组,模型组,金欣高、中、低剂量组及预先给药组,利巴韦林组,每组8只。金欣高、中、低剂量组在大鼠RSV滴鼻3天后分别以0.50g/ml、0.99g/ml、1.98g/ml保留灌肠给药30min,利巴韦林组予以利巴韦林颗粒剂(1g/ml)灌胃。预先给药组在大鼠RSV滴鼻第1天的同时保留灌肠,剂量及操作方法均同金欣中剂量组。各组均每天给药2次,每次1ml/100g,预先给药组连续给药10天,其余各组连续给药7天。测定各组大鼠血清中肿瘤坏死因子(TNF-α)及支气管肺泡灌洗液中分泌型免疫球蛋白A(SIgA)的含量。结果与模型组及金欣低剂量组相比,预先给药组、金欣中剂量组TNF-α水平显著增高(P<0.05或P<0.01);金欣各用药组和利巴韦林组TNF-α水平比较差异无统计学意义(P>0.05)。与模型组及利巴韦林组相比,预先给药组及金欣高、中剂量组的SIgA含量均显著增高(P<0.01);金欣高剂量组SIgA含量亦明显高于预先给药组及金欣中、低剂量组(P<0.05或P<0.01)。结论金欣浓缩液保留灌肠后可能通过增加RSV感染幼年SD模型大鼠的TNF-α、SIgA的分泌量,发挥抗病毒作用,且存在一定的量效关系。Objective To investigate the effect and mechanism of Jinxin Concentrated Solution (Herbal decoction for pneumonia) enema on model rats with pneumonia induced by respiratory syncytial viruses (RSV). Methods Fifty-six SD rats were randomized into seven groups,i.e. the normal control group,model control group,Jinxin high-dosage group,medium group,and low-dosage groups,pretreated group,and Ribavirin group,with 8 in each. The Jinxin high-dosage,medium-dosage,and low-dosage groups were given 0.50g/ml,0.99g/ml,and 1.98g/ml Jinxin Concentrated Solution enema respectively for 30min three days after using RSV nasal drop. The Ribavirin group was given Ribavirin Granule 1g/ml through intragastric administration. The pretreated group was given Jinxin Concentrated Solution enema at the same time of the first day of using RSV nasal drop with the same dosage and manipulation as that of the Jinxin medium-dosage group. All medicated groups were treated twice a day,1ml/100g each time. The pretreated group was treated for 10 days in succession and other groups for 7 days. The level of tumor necrosis factor α (TNF-α) in serum and secretory immuno-globulin A (SIgA) in bronchia alveolus lavage fluid were measured. Results Compared with the model control group and Jinxin low-dosage group,the level of TNF-α in the pretreated group and Jinxin medium-dosage group was significantly higher (P0.05 or P0.01). The level of TNF-α in all Jinxin groups and Ribavirin group was without statistical difference (P0.05). Compared with the model control group and Ribavirin group,the level of SIgA in the pretreated group and Jinxin high-dosage and medium-dosage groups was significantly higher (P0.01). The level of SIgA in the Jinxin high-dosage group was significantly higher than that of the pretreated group and Jinxin medium-dosage and low-dosage groups (P0.05 or P0.01).
