摘 要:依据中心复合旋转设计原理,在单因素试验的基础上,采用三因素五水平的响应曲面分析法,建立了发酵液中多杀菌素提取的二次多项式数学模型,并以多杀菌素质量浓度为响应值作响应面和等高线,考察了提取时间、料液比和提取频率对多杀菌素质量浓度的影响。结果表明:发酵液中多杀菌素超声提取的优化工艺条件为提取频率75 KHz,提取时间74 min,料液比1∶1.5。在此工艺条件下,从发酵液中提取多杀菌素的质量浓度可达到100.75μg/mL。The extraction technique of spinosad from fermentation broth was optimized by central composite rotatable design.On the basis of one-factor experiments,the method of response surface analysis with 3 factors and 5 levels was adopted.A second order equation for extracting spinosad was built.Response surface and contour were graphed with the concentration of spinosad as the response value.Based on the analysis of the response surface plots and their corresponding contour plots,effects of extracting time,the ratio of material to solvent and ultrasonic extracting frequency was explored.The optimal extraction conditions,by using this new method,were determined as follows: extracting frequency 75 KHz,extracting time 74 min and the ratio of material to solvent 1∶1.5.Under the optimized conditions,the average concentration of spinosad was 100.75 μg/mL.
