合欢皮提取物抑制血管生成的体内药效学研究

摘　　要：为了探讨合欢皮提取物体内对血管生成的抑制作用。本文采用MTT法和Matrigel plug方法考察合欢皮提取物对人血管内皮细胞(HMEC)增殖和体内Matrigel胶小管形成的影响,运用Lewis肺癌转移小鼠模型,观察合欢皮提取物对小鼠Lewis肺癌的疗效。结果表明合欢皮提取物可明显抑制HMEC细胞的体外增殖(IC50为30μg/mL),并且呈明显的剂量依赖性,同时具有抑制Matrigel plug中的血管新生的作用。合欢皮提取物还能够明显抑制Lewis肺癌肿瘤细胞生长,减少转移病灶。提示合欢皮提取物具有明显抑制血管生成的作用,有可能成为有效的血管生成抑制剂。To study the inhibition effect of Albizia julibrissin extracts on neovascularization,the effect of A.julibrissin extracts on the proliferation of endothelial cell（HMEC） in vitro and tube formation on Matrigel in vivo were examined by MTT assay and Matrigel plug assay.Therapeutic effect of A.julibrissin extracts on mice with Lewis lung cancer was observed.It was resulted that the proliferation of HMECs was inhibited significantly by A.julibrissin extracts in a dose-dependent manner（IC50 = 30 μg/mL）.It also inhibited angiogenesis in Matrigel plug of mouse model and the growth and metastasis of Lewis lung carcinoma in mice.We found that A.julibrissin extracts,which can inhibit angiogenensis obviously,could be developed as a promising antiangiogenic drug.
