摘 要:黄瓜枯萎病是由半知菌亚门尖孢镰刀菌黄瓜专化型(Fusarium oxysporumf.sp.cucumainumOwen)侵染引起的一种土传病害,是影响黄瓜生产的最主要病害之一。近年来随着分子生物学技术的迅速发展,国内外学者对于病原真菌的遗传多样性做了大量的研究,Wang等[2]对影响黄瓜枯萎病菌AFLP技术体系的多种因素作了探讨,得到了1种适合于黄瓜枯萎病菌AFLP分析的优化体系;Duan等[3]应用RAPD、The genetic diversity of Fusarium specises isolated from cucumber plants showing wilt disease from 8 cities was detected by amplified restriction fragment polymorphism(AFLP).Sixteen combinations of primer pair were selected from 130 combinations and used to amplify the genomic DNA of 351 Fusarium oxysporum isolates.A total of 116 444 bands were obtained and 60 579 of them showed polymorphism,accounting for 52.0%.The genetic distances of 351 F.oxysporum isolates were ranged 0.56-1.00 based on AFLP markers,averaged at 0.78.The results showed that the genetic diversity among the F.oxysporum isolates was high.The 351 tested isolates could be classified into 5 AFLP groups,and most of the isolates from the same area were clustered in the same group by using the UPGMA method.
