摘  要:目的:研究黄芩苷对大鼠原代皮层神经元过氧化氢损伤的保护作用。方法:采用皮层神经元过氧化氢损伤模型研究黄芩苷的抗氧化作用,取不同浓度的黄芩苷溶液,采用DPPH法测定黄芩苷自由基清除能力,用试剂盒测定黄芩苷抗超氧阴离子,通过检测尿酸生成来测定黄芩苷对黄嘌呤氧化酶活性的影响。结果:黄芩苷给药后,过氧化氢损伤后大鼠原代皮层神经元的MTT值升高,上清液中LDH下降、SOD升高,且其可有效清除氧自由基和超氧阴离子及抑制黄嘌呤氧化酶活性,且随着黄芩苷浓度升高,体系中尿酸生成逐渐减少。结论:黄芩苷对过氧化氢损伤的神经元有显著保护作用,该作用可能与上调SOD水平、清除氧自由基和超氧阴离子及抑制黄嘌呤氧化酶活性有关。Objective:To explore the protective effect of baicalin on H2O2 injured rats' primary cortical neurons.Methods:H2O2 was employed to form the injured rats' primary cortical neuron model,and the antioxidative effect of Baicalin was examined by this model.The free radical scavenging effects of different concentrations of baicalin were measured with DPPH method.The superoxide anion resistance detected by kits.The effect of baicalin on xanthine oxidase activity was measured by detecting the content of uric acid produced in xanthine/xanthine oxidase system.Results:Baicalin improved the livability of H2O2 injured rats' primary cortical neurons,superoxide dismutase activity was increased and lactate dehydrogenase was reduced in the neuron culture.Baicalin could scavenge free radical and superoxide anion;in addition,it could inhibit the activity of xanthine oxidase,and the content of uric acid,produced in xanthine/xanthine oxidase system,continuously reduced following increasing baicalin concentrations.Conclusion:Baicalin can exert protective effect on H2O2 injured rats' primary cortical neurons,and this effect could be related to increasing SOD activity,scavenging free radical and superoxide anion directly,and inhibiting xanthine oxidase activity.
