摘  要:目的:比较大花红景天(Radix et Rhizoma Rhodiolae crenulatae)水提剂与红景天苷(Salidroside)对低氧条件下ECV304细胞表达HIF-1α和VEGF的不同影响。方法:细胞分4组:正常氧对照组,低氧对照组,红景天水提剂组,红景天苷组。采用TaqMan探针实时定量聚合酶链式反应(PCR)方法检测mRNA表达水平;Western Blot法检测蛋白表达水平。结果:与正常氧对照相比,低氧促进ECV304细胞HIF-1α和VEGF基因和蛋白表达(P<0.05);同在低氧条件下,红景天水提剂显著促进了ECV304细胞HIF-1α和VEGF的基因和蛋白表达(P<0.01),而红景天苷的促进作用较弱,没有提高低氧时HIF-1α和VEGF的蛋白水平。结论:红景天促进HIF-1α、VEGF表达时,红景天苷不是主要有效成分。Objective:To compare the effect of aqueous extract of Radix et Rhizoma Rhodiolae crenulatae(Rho) and Salidroside on expressions of HIF-1α and VEGF in ECV304 cells under hypoxia conditions.Methods:Four groups were studied:normoxia control group,hypoxia control group,Rho-treated group,and Salidroside-treated group.The mRNA and protein expressions of HIF-1α and VEGF were analyzed respectively by TaqMan real-time quantitative polymerase chain reaction assay(PCR) and Western blot.Results:Both mRNA and protein levels of HIF-1α and VEGF were elevated during hypoxia(P〈0.01) and Rho promoted HIF-1α and VEGF gene and protein expressions remarkably comparing hypoxia control group(P〈0.01).However,the effect of Salidroside was not equal to Rho and had no effect on protein expression of HIF-1α and VEGF during hypoxia.Conclusion:Salidroside was not the main effective component of Radix et Rhizoma Rhodiolae crenulatae in respect of the effect on expressions of HIF-1α and VEGF during hypoxia in vitro.
