用低渗膨胀试验评价黑猩猩冷冻精子的膜完整性

摘　　要：精液品质评定的常见方法有精子活力、畸形率和顶体完整率等指标,但这些指标不能评定精子的真实受精能力。目前低渗膨胀试验作为检验精子膜功能的一种方法得到广泛认可,利用它可检验具有完整膜的精子是否具有生化活性。本试验通过对冷冻2只黑猩猩精液进行低渗膨胀来评定冷冻精子的膜完整性。试验结果显示,本研究中的黑猩猩的冷冻精子在低渗溶液中的膨胀率较高,表明试验黑猩猩的冷冻精液有较高的活力,精子具有较高的穿透率和受精能力。Accepted measures of semen quality include sperm quality,sperm motility,sperm abnormality,acrosome integrity rate,and other indicators.However,these indicators cannot accurately assess fertilization ability.HOST（Hypoosmotic swelling test） tests sperm membrane function and is currently widely recognized as a useful test of sperm membrane biological activity.This study we used the low permeability expanding test to assess the integrity of sperm membranes of frozen sperm from two captive Chimpanzees at Guangzhou Zoological Garden.The expansion rate of frozen sperm in hypotonic solution was high,indicating a high percentage of the frozen Chimpanzee sperm cells were alive,sperm cells were capable of penetration, and therefore were likely to have good fertilization ability.
