摘 要:用SPF鸡胚从2005-2006年采集自天津地区猪群样品中分离出3株猪流感病毒,测定了分离病毒的部分理化特性和生物学特性。3个猪流感病毒分离株均能凝集1%鸡红血球,将其制成琼脂扩散抗原与猪流感标准阳性血清反应均出现清晰白色沉淀线;经中国动物卫生与流行病学中心系统鉴定,3个毒株均为H1N1亚型,分别命名为A/Swine/Tianjin/TJ2/2005(H1N1)、A/Swine/Tianjin/TJ3/2005(H1N1)、A/Swine/Tianjin/TJ4/2006(H1N1)株。这3株病毒都为热不稳定型,60℃作用10 min即可使病毒失去感染能力,病毒对酸性环境(pH3.0)和乙醚均敏感。TJ2、TJ3、TJ4株的EID50依次为10-7.5/0.2 mL、10-7.17/0.2 mL和10-8.45/0.2 mL。人工感染试验结果表明,分离的3株病毒对鸡和小鼠无致病性,但对猪具有致病性,能引起部分感染猪发病,并产生明显的肺部病理变化,从发病猪体内均能分离到H1N1亚型猪流感病毒。Three strains of swine influenza virus(SIV)were isolated from pig farms in Tianjin region in 2005 to 2006.All of the three isolates showed hemagllutinin activity to 1% RBC of cock and were positive in AGP test with antibody of SIV.Systemic identification was done by China Animal Healthy and Epidemiology Center and the result showed the three isolates were belong to H1N1 subtype.The isolates were christened as A/Swine/Tianjin/TJ2/2005(H1N1),A/Swine/Tianjin/TJ3/2005(H1N1)and A/Swine/Tianjin/TJ4/2006(H1N1).All of the isolates of SIV were became uninfected to chicken embryo in 60℃ water bath 10 min,pH3.0 incubated 3 h at 25℃ and incubated overnight in 4℃ with aether.EID50 of the three isolates were 10-7.5/0.2 mL,10-7.17/0.2 mL and 10-8.45/0.2 mL respectively.The isolates of SIV couldn′t cause disease in chicken and mice after infected,but some of pigs were get sick post inoculated in all group infected by the three isolates and H1N1 subtype of SIV were gotten from lungs of infected pigs.
