摘 要:采集成年健康母猪的卵巢,收集卵泡颗粒细胞,在配制好的培养液中培养。颗粒细胞原代培养体系建立成功后,分别将不同质量浓度的FSH(0.1,0.5,1,5,25 ng/mL)作用于细胞,每隔48 h换1次培养液,加入新的FSH,倒置显微镜下检测细胞的生长及增殖状况。结果表明,FSH质量浓度越大,细胞生长状况越好,差异有统计学意义(P<0.05);在固定质量浓度的FSH作用下,细胞每次换液后生长状况都会变好。FSH能促进猪卵泡颗粒细胞增殖,并与作用时间长短有关。To build the primary system of ovarian granulose cells in pigs, ripe female pig's ovarian granulose cells were collected and cultured in the prepared culture medium. After pig ovarian granulose cells were prepared, cells were exposed to different densities of 0.1,0.5,1,5,25 ng/mL FSH respectively, and the culture medium was changed every 48 hours and new FSH added. Growth and proliferation rate were analyzed by inverted microscope. The result showed that the higher the density of FSH, the better the growth and propagate condition were(P〈O.05 ). If it was under fixed density of FSH, the growth and proliferation rate increased significantly after change the culture medium every 48 hours. Conclusion was drawn as FSH encouraged the growth of ovarian granulose cells in pigs and it might be related with the action time.
