穴位敷贴对过敏性鼻炎小鼠脾淋巴细胞特异性增殖的影响

摘　　要：【目的】观察穴位敷贴对过敏性鼻炎小鼠脾淋巴细胞特异性增殖的影响。【方法】选用BALB/c小鼠,采用卵白蛋白(OVA)腹腔注射致敏加滴鼻法复制过敏性鼻炎模型,造模成功后小鼠随机分为穴位敷贴组(中药敷贴大椎穴)、假治疗组(纸团代替药物敷贴大椎穴)、地塞米松组(剂量为0.05 mg/只)和模型组,并设立正常对照组;连续治疗4周后,分离培养各组小鼠脾T淋巴细胞,并分别以10μg/mL OVA体外刺激3 d,采用四甲基偶氮唑盐(MTT)法测定各组细胞的增殖活性。【结果】模型组与假治疗组增殖指数显著升高,与正常对照组比较差异有显著性意义(P<0.001),而假治疗组与模型组比较差异无显著性意义(P>0.05);穴位敷贴组与地塞米松组均可降低增殖指数,与模型组比较差异有显著性意义(P<0.001)。【结论】穴位敷贴治疗过敏性鼻炎的作用与其能抑制脾淋巴细胞免疫反应有关。Objective To observe the effect of acupoints application of herbal medicine on specific proliferation of spleen lymphocytes in allergic rhinitis mice.Methods BALB/c mice were used as the experimental animal.The allergic rhinitis models were induced by intraperitoneal injection and nasal dripping of oval albumin（OVA）,and then the model mice were randomized into acupoints application group（external application of herbal medicine on Dazhui point）,pseudo-acupoints application group（external application of paper ball on Dazhui point）,dexamethasone group（in the dosage of 0.05 mg for each mouse）,and model group.The normal control group was also set up.After treatment for 4 continuous weeks,T lymphocytes were isolated from the mice spleen,and then cultured with OVA 10 μg/mL for 3 days.Methyl thiazolyl tetrazolium（MTT） colorimetry was used for the determination of proliferation of T lymphocytes.Results The proliferation index was increased in the model group and pseudo-acupoints application group（P〈0.001 compared with that in the normal control group）,and the difference was insignificant between the two groups（P〉0.05）.In acupoints application group and dexamethasone group,the proliferation index was decreased（P〈0.001 compared with that in the model group）.Conclusion The therapeutic mechanism of acupoints application of herbal medicine for allergic rhinitis is probably related with the inhibition of immune reaction of spleen lymphocytes in mice
