摘 要:【目的】观察滋肾宁神胶囊对D-半乳糖所致亚急性衰老模型大鼠的影响。【方法】选用SD大鼠随机分成空白对照组,模型组,七宝美髯丸组(剂量为2 g.kg-1.d-1),滋肾宁神丸组(剂量为3.33 g.kg-1.d-1),滋肾宁神胶囊高、中、低剂量组(剂量分别为9.96、4.98、2.49 g.kg-1.d-1);除空白对照组外,其他组均采用D-半乳糖(剂量为200 mg.kg-1.d-1)腹腔注射30 d复制亚急性衰老模型,第31天开始各组分别按设计剂量连续给药90 d。取各组大鼠大脑皮层检测超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性,丙二醛(MDA)含量,以及神经递质去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)的含量。【结果】模型组大鼠SOD、GSH-PX活力及NE、DA、5-HT含量均下降,MDA含量升高,与空白对照组比较差异具有显著性意义(P<0.05或P<0.01);滋肾宁神胶囊高、中剂量组可升高SOD、GSH-PX活力和NE、DA含量,降低MDA含量,低剂量组可升高SOD、GSH-PX活力和NE含量,与模型组比较差异均有显著性意义(P<0.05或P<0.01)。【结论】滋肾宁神胶囊抗衰老作用与其能提高SOD、GSH-PX活力,增强清除自由基能力,提高单胺类神经递质含量有关。Objective To investigate the anti-aging effect of Zishen Ningshen Capsules(ZNC) on rat models of sub-acute aging induced by D-galactose.Methods SD rats were randomized into blank control group,model group,Zishen Ningshen Pills group(in the dosage of 3.33 g·kg^-1·d^-1and high-,middle-and low-dosage ZNC groups(in the dosages of 9.96,4.98 and 2.49 g·kg^-1·d^-1espectively).Except for the blank control group,the rats in the other groups were given intraperitoneal injection of D-galactose(200 mg·kg^-1·d^-1for 30 days to induce sub-acute aging.On the 31st day of the experiment,the rats were medicated with the corresponding drugs according to the experimental design.After treatment,we detected superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX) activities,malondialdehyde(MDA) content,and the levels of neurotransmitters of norepinephrine(NE),dopamine(DA),5-hydroxytryptamine(5-HT)in rats cerebral cortex.Results In the model group,SOD and GSH-PX activities,and NE,DA and 5-HT levels were decreased,and MDA content was increased(P〈0.05 or P〈0.01 compared with those in the blank control group).SOD and GSH-PX activities and NE and DA levels were increased,and MDA content was decreased in high-and middle-dosage ZNC groups.Low-dosage ZNC had an effect on increasing SOD and GSH-PX activities and NE level(P〈0.05 or P〈0.01 compared with those in the model group).Conclusion The anti-aging mechanism of ZNC is probably related with the increase of SOD and GSH-PX activities,free radical clearance and neurotransmitters levels in rat models of sub-acute aging induced by D-galactose
