摘　　要：以黄萎病菌（从邯208中分离到的致病性中等的和从中棉所8号中分离到的致病性弱的菌株）为材料,构建了AD-cDNA文库。该文库的转化效率为4×105cfu/μg,库容量为4.8×106克隆,插入片段的大小在750~1 500 bp之间。筛选获得的候选蛋白测序结果表明含有TRRAP结构域（TRansformation/tRanscription domain-Associated Protein,转化/转录域相关蛋白）,推测TRRAP是一个锚定蛋白,是Avr蛋白和R蛋白相互识别和交换信号的场所,其功能和作用机制有待进一步的研究。该文库可以用于棉花抗黄萎病的相关研究,并为进一步解释棉花抗黄萎病菌的机理提供研究基础。An AD-cDNA library of Verticillium dahliae isolated from Han 208 and Zhongmiansuo NO.8 was constructed.The library＇s transformation efficiency was 4×105cfu/μg with the cloning capacity of 4.8×106 recombinants,and the length of insertion fragment was from 750 bp to 1 500 bp.The sequence results of positive colonies showed that they were Transformation/Transcription domain-Associated Proteins.TRRAP was inferred to be a docking protein,in which Avr protein and R protein proceeded the mutual recognition and signaling.This library could be used for the research related to cotton Verticillium wilt resistance,and would be aid to explain its mechanism.
