大肠癌细胞对化疗药物的体外敏感性研究

摘　　要：目的利用噻唑蓝(MTT)比色法探讨大肠癌细胞对肿瘤化疗药物的体外敏感性。方法对36例患者的大肠癌组织进行体外培养,利用MTT法测定6种临床常用化疗药物的抑制率。结果 36例大肠癌试验中,化疗药物的敏感性从高到低依次为丝裂霉素(MMC)、顺铂(DDP)、5-氟尿嘧啶(5-FU)、依托泊苷(VP-16)、奥沙利铂(L-OHB)、阿霉素(ADM)。其中18例结肠癌试验中,化疗药物的敏感性从高到低依次为MMC,DDP,5-FU,VP-16,L-OHB,ADM;18例直肠癌试验中,化疗药物的敏感性从高到低依次为MMC,DDP,5-FU,L-O-HB,VP-16,ADM。结论对于大肠癌,MMC,DDP,5-FU的敏感性较高,可作为首选药物;对于结肠癌和直肠癌,也有相似的规律性。Objective To study the in vitro sensitivity of colorectal cancer cells to chemotherapeutic drugs by MTT assay.Methods The colorectal cancer tissues from 36 cases were cultured in vitro.The inhibition rate to cancer cells for selected 6 kinds of commonly used chemotherapeutic drugs in clinic were determined by MTT assay.Results In 36 cases of colorectal cancer,the sensitivity of those 6 drugs from high to low was MMC,DDP,5-FU,VP-16,L-OHB and ADM.Among 18 cases of colonic cancer,the sensitivity sequence of those 6 drugs from high to low was MMC,DDP,5-FU,VP-16,L-OHB and ADM.Among 18 cases of rectal cancer,the sensitivity sequence of those 6 drugs from high to low was MMC,DDP,5-FU,L-OHB,VP-16 and ADM.Conclusion MMC,DDP and 5-Fu have highly sensitive to colorectal cancer,which can be used as the first choice of drugs.Colonic and rectal cancers have the similar regularity in sensitivity of chemotherapeutic drugs.
