摘  要:目的探讨阿托伐他汀对动脉粥样硬化(Atherosclevosis,AS)兔血管壁中过氧化物酶增殖激活受体γ((peroxi-some proliferator activated receptorγ,PPAR-γ)表达的影响。方法雄性新西兰大白兔44只采用随机数字表法分为对照组(CON组)、模型组(AS组)、低剂量组(LS组)和高剂量组(HS组),每组11只。适应性喂养5天后,AS、LS、HS组作主动脉球囊损伤,术后第1天即开始予以下方案喂养:AS组予高脂饲料120 g/d喂养,LS组予高脂饲料120 g/d+阿托伐他汀2.5 mg/d,HS组予高脂饲料120 g/d+阿托伐他汀5.0 mg/d;CON组仅作单侧股动脉结扎,术后予以普通颗粒饲料120 g/d喂养。饲养14周后44只全部处死,获取腹腔干以下2.5 cm处的腹主动脉段,HE染色作常规病理分析,并用免疫组织化学方法(SP法)检测PPAR-γ在血管壁的表达。结果经"球囊损伤+高脂饮食"处理的3组有不同程度的斑块形成,PPAR-γ在血管壁的表达明显增加(P<0.01),并且与阿托伐他汀的剂量有关;与AS组比较,LS组和HS组血管壁PPAR-γ表达进一步增加(P<0.01);HS组与LS组比较,PPAR-γ表达的阳性细胞率进一步增加(P<0.01)。结论 "球囊损伤+高脂饮食"可诱导兔主动脉粥样硬化模型形成;AS的形成可引起PPAR-γ表达适应性增强;阿托伐他汀干预可进一步增强PPAR-γ的表达,并且与剂量有关。Objective To explore the effect of Atorvastatin on the expression of peroxisome Proliferator-activated receptor-γ(PPAR-γ)in vessel wall of rabbits with atherosclerosis.Methods Forty-four New Zealand male rabbits were randomly divided into control group(CON),atherosclerosis model group(AS),low-dose Atorvastatin group(LS)and high-dose Atorvastatin group(HS),with 11 members in each group.The AS group,LS group and HS group got balloon injury after feeding adaptively for 5 days.And then they began their feeding from the first day after operation according to the following suggestions:the AS group was fed by high-fat fodder of 120 g/d,LS group by high-fat fodder of 120 g/d and atorvastatin of 2.5 mg/d,HS group by high-fat fodder of 120 g/d and Atorvastatin 5.0 mg/d.The CON group,after single femoral artery ligature,was fed on ordinary pelleted fodder of 120 g/d.Fourteen weeks later,all the rabbits were killed in order to get abdominal aorta which was 2.5 cm under celiac trunk.After routine pathological analysis of HE staining,the PPAR-γ expression on vessel wall was tested by immunohistochemistry methods(SP).Results Plaque existed in certain degrees in AS,LS and HS group with aortic balloon injury and high-fat diet and meanwhile the rate of positive cell of PPAR-γ expression increased(P0.01).Compared with AS group,the PPAR-γ expression on vessel wall for LS and HS group had further increased the rate of positive cell(P0.01).In contrast to LS group,positive cell of PPAR-γ expression of HS group had still kept rising(P0.01).For the three groups with ortic ballon injury and high-fat diet,the PPAR-γ expression on vessel wall was abviously increased,which was associated with the dose of Atorvastatin.Conclusions Balloon injury and high-fat diet can induce the forming of aortic rabbit atherosclerosis model.The forming of AS can enhance the expressivity of PPAR-γ adaptivity.The intervention of atorvastatin can further enhance the expression of PPAR-γ which is associated with the d
