摘　　要：目的　建立一种灵敏度高、特异性好的 Ｄ Ｎ Ａ 探针检测葡萄球菌肠毒素 Ｂ 基因方法。方法　用聚合酶链反应（ Ｐ Ｃ Ｒ） 扩增的方法进行葡萄球菌 Ｂ 型肠毒素基因探针生物素标记，用制备好的基因探针，进行斑点杂交，对葡萄球菌肠毒素１０ 株标准株和３９ 株临床株分别进行了检测。结果　探针灵敏度为２５０ ｐｇ ，检测模板灵敏度为０９８ ｎｇ 。对西安、济南、北京等地临床分离株，进行 Ｐ Ｃ Ｒ 和斑点杂交方法的比较，阳性符合率达到１００ ％ 。结论　用 Ｐ Ｃ Ｒ 扩增的方法标记生物素基因探针，建立斑点杂交的方法，为葡萄球菌肠毒素 Ｂ 基因的诊断提供了科学依据。Objective To establish a sensitive and specific gene probe method for detection of Staphylococcal enterotoxin B(SEB) gene. Methods Gene probe labeled with biotin by PCR was detected by dot hybridization, and separately 10 standard strains and 39 clinical specimens were detected Results The sensitivity of gene probe was 0 25 ng The limit of method was 0 98 ng The dot hybridization method compared with PCR method on detection of clinical specimens from Xian, Jinan, and Beijing The accordance rate of positivity was 100% Conclusion The dot hybridization with biotin labeling gene probe by PCR provides an etiological basis for the diagnosis of SEB gene
