摘 要:采用不同浓度尿素和盐酸胍从小牛骨基质中分离纯化出牛骨形成蛋白(bBMP) ,将其植入小鼠股部的肌肉内观察诱导成骨过程。组织学和酶组织化学研究表明 ,bBMP诱导间充质细胞逐渐分化形成软骨细胞和成骨细胞 ,最终新骨形成。诱导成骨的时间和过程与以往报道一致。bBMP诱导骨形成过程呈现连续又具显著的阶段性。本研究结合组织学和酶组织化学观察的特点 ,将其全过程分为4个时期 ,(1)间充质细胞增生期 ;(2)软骨细胞分化期 ;(3)新骨组织形成期 ;(4)骨组织成熟期。Bovine bone morphogenetic protein (bBMP) was separated and purified from bovine bone by repeated solubilization and precipitation of the protein with different concentration of Urea and GuHCI according to the Yang's method. The property of inductive osteogenesis of bBMP caused new bone formation in mouse thigh muscles. A gradual transformation of mesenchymal-type cells into active chondroblast and osteoblasts and finally into heterotopic new bone was observed and studied histologically and enzymehistochemically. The results showed the process of inductive bone formation was much the same trem with in the time and sequences as it previously reported. The whole process of the morphogenetic response occurred in this study was divided into four stages according to the histological and enzymehistochemical characteristics. In order of occurrence, the histogenic responses are (1)mesenchymzal cell activation and proliferation;(2) cartilage formation; (3) bone formation ;(4) lamellar bone formation.
