摘 要:目的:观察心肌细胞织反常时细胞内的游离钙浓度的变化,探讨丹参素和SOD对细胞内的游离钙浓度的影响。方法:体外分离成年大鼠心肌细胞复制氧反常模型,缺氧40min后给氧20min,用钙荧光探针(Fluo-3)分别测定了正常大鼠心肌细胞(Control,n=11)、缺氧40min(H40min,n=11)以及缺氧40min再给氧20min(H40minR20min,n=12)的大鼠心肌细胞内的游局钙浓度,并观察了丹参素(H40minR20min+DS,n=10)和SOD(H40minR20min+SOD,n=11)对氧反常的心肌细胞内的游离钙浓度的影响,并用显微镜观察了细胞杆形率的改变。结果:氧反常组的细胞内的游离钙浓度(596.71±20.29nmol/L)明显比正常对照和单纯缺氧40min的高(158.67±5.01和328,51±13.86nmol/LP<0.01)。加入丹参素和SOD,可明显降低氧反常细胞内的游离钙的浓度(分别为245.29±8.83和300.04±13.04nmol/L,P<0.01)。细胞杆形率亦有类似的改变。结论:氧反常时心肌细胞发生明显的钙超载,丹参素和SOD对细胞内钙超载具有一定的防治作用。To observe the changs of intracellular free calcium concentration in oxygen-paradoxed rat myocardial cells and to discuss the proective efforts of Salvia Miltiorrhizae (DS) and SOD on Ca2+ - overloaded rat myocardialcells. METHODS: Adult rat myocardial cells was isolated in vitro and oxygen paradox model was made. Selecting 40 minas hypoxia period followed by 20 min of reoxygenation, a new calcium fluoresence probe (Fluo - 3) was used to detect theintracellular free calcium concentration in control (n = 11) 、 H40min(hypoxia, n = 11 ) and H40minR20min (hypoxia+reperfuxion, n = 12) groups respectively. We also looked into the rod-shaped rates of myocardial cells with microscope.Inaddition, the effect of Salvia Miltiorrhizae (n = 10) and SOD (n = 11 ) on intracellular free calcium concentration and rod-shaped rate of oxygen-paradoxed myocedal cells were observed. RESULTS: The intracellular free calcium concentrationof H40minR20min group increased far beyond the control and H40min groups(596. 71 ± 20. 29 nmol/L vs 158. 67 ± 5. 01and 328.51 ± 13 .86 nmol/L, P < 0.01 ), showing a significant Ca2+ - overload. The changes of rod-shaped rates weresimilar to intracellular free calcium concentration. Salvia Miltiorrhizae and SOD can inhibit Ca2+ - overload obviously (254.29 ± 8. 83 and 300.04± 13 .04 nmol/L vs H40minR20min 596. 71 ± 20. 29 nmol/L, P < 0.01). Moreover, effect of Salvi-a Miltiorrhizae was more striking than SOD (P < 0.01 ). CONCLUSION: There is a significant Ca2+-overload in oxygen- peoxed myocedal cells. Salvia miltiorrhizae and SOD have a definite protective effect on Ca2+ - overload.
